Le laboratoire, créé en 1969, porte le nom de son fondateur Jacques-Louis Lions. il s'agit maintenant d'une unité de recherche conjointe à l’Université Pierre et Marie Curie, à l’université Paris Diderot et au Centre National de la Recherche Scientifique.
Le Laboratoire Jacques-Louis Lions constitue le plus grand laboratoire de France et l'un des principaux au monde pour la formation et la recherche en mathématiques appliquées.
Il accueille l'activités de deux masters deuxième année ce qui représente un centaine d'étudiants. Ses activités recouvrent l’analyse, la modélisation et le calcul scientifique haute performance de phénomènes représentés par des équations aux dérivées partielles.
Fort d’environ 100 enseignants-chercheurs, chercheurs, ingénieurs, personnels administratifs permanents ou émérites, et d’autant de doctorants ou post-doctorants, il collabore avec le monde économique et avec d'autres domaines scientifiques à travers un large spectre d'applications : dynamique des fluides; physique, mécanique et chimie théoriques; contrôle, optimisation et finance; médecine et biologie; traitement du signal et des données.

Desc: Self-assembly of proteins into amyloid aggregates is an important biological phenomenon associated with human diseases such as Alzheimer's disease. Amyloid fibrils also have potential applications in nano-engineering of biomaterials. The kinetics of amyloid assembly show an exponential growth phase preceded by a lag phase, variable in duration as seen in bulk experiments and experiments that mimic the small volumes of cells. Here, to investigate the origins and the properties of the observed variability in the lag phase of amyloid assembly currently not accounted for by deterministic nucleation dependent mechanisms, we formulate a new stochastic minimal model that is capable of describing the characteristics of amyloid growth curves despite its simplicity. We then solve the stochastic differential equations of our model and give mathematical proof of a central limit theorem for the sample growth trajectories of the nucleated aggregation process. These results give an asymptotic description for our simple model, from which closed form analytical results capable of describing and predicting the variability of nucleated amyloid assembly were derived. We also demonstrate the application of our results to inform experiments in a conceptually friendly and clear fashion. Our model offers a new perspective and paves the way for a new and efficient approach on extracting vital information regarding the key initial events of amyloid formation.

Desc: Les vibrations non linéaires géométriques de coques minces, discrétisées par éléments finis sont considérées. Dans le but de réduire la dynamique vibratoire à des modèles comprenant un petit nombre de degrés de liberté, la base des modes propres linéaires est utilisée. La projection fait alors apparaitre un ensemble de coefficients de couplage non linéaires quadratiques et cubiques. Une méthode directe permettant le calcul de ces coefficients est proposée et implémentée. Une fois le modèle projeté, des études de bifurcation de solutions périodiques sont possibles a moindre coût, ce qui permet de calculer les branches stables et instables des solutions libres et forcées.

Desc: Single ventricle heart defects are among the most serious congenital heart diseases, and are uniformly fatal if left untreated. Typically, a three-staged surgical course, consisting of the Norwood, Glenn, and Fontan surgeries is performed, after which the superior vena cava (SVC) and inferior vena cava (IVC) are directly connected to the pulmonary arteries (PA). In an attempt to improve hemodynamic performance and hepatic flow dis- tribution (HFD) of Fontan patients, a novel Y-shaped graft has recently been proposed to replace the traditional tube-shaped extracardiac grafts. Previous studies have demon- strated that the Y-graft is a promising design with the potential to reduce energy loss and improve HFD. However these studies also found suboptimal Y-graft performance in some patient models. The goal of this work is to determine whether performance can be improved in these models through further design optimization. Geometric and hemody- namic factors that influence the HFD have not been sufficiently investigated in previous work, particularly for the Y-graft. In this work, we couple Lagrangian particle tracking to an optimal design framework to study the effects of boundary conditions and geometry on HFD. Specifically, we investigate the potential of using a Y-graft design with unequal branch diameters to improve hepatic distribution under a highly uneven RPA/LPA flow split. As expected, the resulting optimal Y-graft geometry largely depends on the pulmo- nary flow split for a particular patient. The unequal branch design is demonstrated to be unnecessary under most conditions, as it is possible to achieve the same or better per- formance with equal-sized branches. Two patient-specific examples show that optimization-derived Y-grafts effectively improve the HFD, compared to initial nonopti- mized designs using equal branch diameters. An instance of constrained optimization shows that energy efficiency slightly increases with increasing branch size for the Y-graft, but that a smaller branch size is preferred when a proximal anastomosis is needed to achieve optimal HFD.

Desc: Self-assembly of proteins is a biological phenomenon which gives rise to spontaneous formation of amyloid fibrils or polymers. The starting point of this phase, called nucleation exhibits an important variability among replicated experiments.To analyse the stochastic nature of this phenomenon, one of the simplest models considers two populations of chemical components: monomers and polymerised monomers. Initially there are only monomers. There are two reactions for the polymerization of a monomer: either two monomers collide to combine into two polymerised monomers or a monomer is polymerised after the encounter of a polymerised monomer. It turns out that this simple model does not explain completely the variability observed in the experiments. This paper investigates extensions of this model to take into account other mechanisms of the polymerization process that may have impact an impact on fluctuations.The first variant consists in introducing a preliminary conformation step to take into account the biological fact that, before being polymerised, a monomer has two states, regular or misfolded. Only misfolded monomers can be polymerised so that the fluctuations of the number of misfolded monomers can be also a source of variability of the number of polymerised monomers. The second variant represents the reaction rate $\alpha$ of spontaneous formation of a polymer as of the order of $N^{-\nu}$, with $\nu$ some positive constant. First and second order results for the starting instant of nucleation are derived from these limit theorems. The proofs of the results rely on a study of a stochastic averaging principle for a model related to an Ehrenfest urn model, and also on a scaling analysis of a population model.

Desc: L'objet de cette thèse est l'étude et la mise en œuvre d'une méthode d’éléments finis multi-échelles pour la simulation d'écoulements miscibles en milieux poreux. La définition des fonctions de base multi-échelles suit l'idée introduite par F. Ouaki. La nouveauté de ce travail consiste à combiner cette approche multi-échelle avec des éléments finis de type Galerkine Discontinus (DG) de façon à pouvoir utiliser ces nouveaux éléments sur des maillages non-conformes composés de mailles de formes diverses. Nous rappelons, dans un premier temps, le principe des méthodes DG et montrons comment ces méthodes peuvent être utilisées pour discrétiser une équation de convection-diffusion instationnaire identique à celle rencontrée dans le problème d'écoulement considéré dans ce travail. Après avoir vérifié l'existence et l'unicité d'une solution à ce problème, nous redémontrons la convergence des méthodes DG vers cette solution en établissant une estimation d'erreur a priori. Nous introduisons, ensuite, les éléments finis multi-échelles non conformes et détaillons leur mise en œuvre sur ce problème de convection-diffusion. En supposant les conditions aux limites et les paramètres du problème périodiques, nous montrons une nouvelle estimation d'erreur a priori pour cette méthode. Dans une seconde partie, nous considérons le problème d'écoulement complet où l'équation considérée dans la première partie est résolue de manière couplée avec l'équation de Darcy. Nous introduisons différents cas tests inspirés de modèles d'écoulements rencontrés en géosciences et comparons les solutions obtenues avec les deux méthodes DG, à savoir la méthode classique utilisant un seul maillage et la méthode étudiée ici. Nous proposons de nouvelles conditions aux limites pour la résolution des problèmes de cellule qui permettent, par rapport à des conditions aux limites linéaires plus classiquement utilisées, de mieux reproduire les variations des solutions le long des interfaces du maillage grossier. Les résultats de ces tests montrent que la méthode multi-échelle proposée permet de calculer des solutions proches de celles obtenues avec la méthode DG sur un seul maillage et de réduire, de façon significative, la taille du système linéaire à résoudre à chaque pas de temps.
